CCAM Microscopy Facility

The CCAM Microscopy Facility is open to the UCHC research community as well as other academic and industrial institutes. Equipment may be used by authorized users who have completed the required training for each piece of equipment they choose to use. Alternatively, CCAM staff are available for service work at an hourly rate. CCAM offers 25Gb of storage space to each lab with an active account.

Confocals

• Zeiss LSM 510/Confocor II combined confocal/NLO/FCS system, mounted on an Axiovert 200, equipped with an argon ion laser with emission at 488, 477, 514, and 543 and with a Coherent Mira femtosecond T-Sapphire laser for nonlinear optical microscopy (2-photon) which is capable of 700-1000nm single optics set tuning.
• Zeiss LSM 510 confocal microscope mounted on an Axiovert 100M, with automated xyz control. Lasers include an argon laser with emission at 364 nm, an argon laser with emission at 458, 488, and 514nm, and two HeNe lasers with emission wavelengths at 543 and 633 nm. The system has 3 photomultiplier tubes and a transmitted light detector.
• Zeiss LSM 510 Meta confocal microscope mounted on an Axiovert 200M with automated xyz control. Laser excitation lines include 458, 488, and 514nm (argon laser) and 543 and 633 nm (HeNe lasers). The system is equipped with the specialized Meta detector as well as 2 traditional photomultiplier tubes and a transmitted light detector. This system is also equipped with the eppendorf InjectMan NI 2 microinjection set.
• Zeiss LSM 410 confocal microscope mounted on an Axiovert 135. Laser is an argon-krypton laser with emission at 488, 568, and 647 nm and a transmitted light detector. The system allows for simultaneous collection of two fluorescence signals and a transmitted image.
• Perkin Elmer UltraView Spinning Disk Confocal

Widefield Microscope

• Wide-field fluorescence imaging system utilizing a Zeiss Axioplan 2 Imaging microscope, equipped with a Photometrics PXL-EEV37 high speed digital cooled CCD camera. Molecular Devices MetaMorph, for image acquisition, analysis and image processing. Basic filter sets include Rhodamine, FITC and DAPI. Equipped for DIC and phase contrast.
• Zeiss Total Internal Reflection Fluorescence (TIRF) imaging system including 100 mW argon laser with 458, 488 and 514 nm excitation, Axio Observer fully motorize microscope and Axiocam MRM camera controlled by AxioVision software.

Software and Additional Equipment

• ImarisSurpass 3-D rendering and analysis software from Bitplane.
• Molecular Devices' MetaMorph;software for analysis, image processing as well as acquisition and device control.
• Canon CanoScan 8400F flat bed scanner
• Hitachi's R-1200 Rapid Scan 60Mhz permanent magnet NMR Spectrometer
• Spex Fluorolog dual excitation, dual emission spectrofluorimeter.
• Additional equipment and facilities are also available in the laboratories of the CCAM faculty.
